Sequence-selective Dna Methylation Analysis Induced by Bulge Specific Immuno-recognition on a Surface Plasmon Resonance Fluidic Chip

This paper reports the sequence-selective immunoassay chip for DNA methylation analysis by microfluidic based surface plasmon resonance (SPR) detection. This was achieved by utilizing an affinity measurement involving the target methylcytosine in a bulge region and anti-methylcytosine antibody in a microchannel, following hybridization with a biotinylated bulge-inducing DNA probe. We obtained the methylation status in 45 min without employing the conventional bisulfite reaction, PCR amplification or sequencing.
